CWRU is seeking a Cryo-EM manager for its new state-of-the-art EM facility. The Cryo-EM manager will assist in the installation and support the operation of the new Titan Krios G3i equipped with a K3 Direct Electron Detector and Gatan Bioquantum energy filter for single particle analysis and cryo-tomography studies. The Titan Krios is also equipped with a phase plate, which enables imaging and reconstruction of relatively small biomolecules. The Cryo-EM manager will work under the supervision of the Faculty Director and oversee the Cryo-EM core’s day-to-day operations and assist CWRU’s technology development efforts.
· User Training - The Cryo-EM manager will train users (undergraduate students, graduate students, post-docs, and Faculty) in sample preparation and imaging techniques for single-particle cryo-EM. Provide guidance and troubleshoot cryo-EM experiments. Will provide advice on data processing.
· Cryo-EM Service – Support local and external users in sample preparation for single particle cryo-EM, facilitate preliminary screening of samples via negative staining techniques, optimize freezing conditions, and collect images. The cryo-EM manager will provide feedback on the quality of the data obtained by the users. Implement data collection strategies. Maintain data collection computers, implement EM-related IT resources, validate and bench new EM software releases. Interface with our High Performance Computing team, who will assist with the computational needs of the users for optimal data storage and data processing.
· Cryo-EM Equipment Maintenance – The cryo-EM manager will be responsible for the alignment of electron microscopes, general maintenance of the Titan Krios cryo-EM microscope and ancillary cryo-EM equipment such as FEI Vitrobot. Will periodically liaise with the manufacturer (FEI/ThermoFisher) regarding preventative maintenance and repairs. Will be responsible for scheduling microscope through iLab, maintain records of equipment usage, report maintenance issues of equipment and schedule repairs, prepare monthly invoices for user charges and prepare monthly status report for the Faculty Director. Will ensure that the preparatory areas and the microscopy rooms are clean and in compliance with EHS protocols and procedures.
Experience: 5-7 years of research experience in a laboratory setting with 2-3 years of cryo-EM experience.
Education/Licensing: PhD degree in Structural Biology or related field.
Demonstrated skill and extensive experience in the high-resolution operation of cryo-electron microscopes. Expertise in sample freezing, sample handling, microscope use, and data evaluation and analysis using EM software (e.g. EMAN, Chimera, Relion, Frealign) is essential. Familiarity with structure determination/analysis workflows and strong knowledge of Linux and OSX operating systems is required. Proven ability to contribute to experimental design, and assist with the analysis and interpretation of results. Strong communication and interpersonal skills are required.
