The responsibilities of this position include planning and executing research projects within a laboratory focusing on cancer drug development. The individual will work closely with multi-investigator team consisting of clinicians and basic scientists within the Case GI Specialized Program of Research Excellence (SPORE) and the National Center for Regenerative Medicine (NCRM). The individual will lead a specific research team consisting of a graduate student and research assistant within the group. This individual will focus on research involving drug development of small molecule inhibitors of 15-PGDH for use in inflammatory bowel disease and bone marrow transplantation. Specific experimental studies will require ability to produce recombinant 15-PGDH protein, to measure PGE2 production in cell line and tissue samples using Elisa assays, to synthesize tritium labeled PGE2 and use it to determine inhibition of 15-PGDH enzyme activity in in vivo studies, to measure 15-PGDH enzyme activity using fluorescent assays for determining IC50 of enzyme inhibitors, to administer 15-PGDH inhibitors to mice IV, IP, and PO, and to harvest tissues for analysis of PGE2 level and 15-PGDH activity. General skills in protein chemistry, enzymology, and animal studies will be required. The individual will be responsible for managing and designing individual experiments, reporting these in a consistent fashion and in a timely manner, managing issues of the scientific method including positive and negative controls, reproducibility, sample size estimate valuation, trends and troubleshooting, and refining techniques into standard operating procedures in the laboratory. Careful documentation and accountability is essential. The individual will be responsible for writing and submitting manuscripts reporting research results.
- Assay in vivo and in vitro activity of 15-PGDH inhibitor compounds (30%).
- In vivo murine techniques including IP, IV, PO drug administration and tissue harvest. In vitro techniques including qRT-PCR, cell culture, Western blot, cell growth, production of recombinant proteins, radiolabeling of ligands in enzymatic reactions, ELISA (30%).
- Manage research team consisting of graduate student and research assistant. (15%)
- Management of research collaboration between Case GI SPORE and National Center for Regenerative Medicine. (5%)
- Preparation and submission of publications of research and preparation and submission of mentored research awards in collaboration with the PI. (15%)
- Write Standard Operating Procedures (SOPs) and maintain detailed laboratory records. (5%)
- Presentation of scientific findings at internal, external, and national meetings. (5%)
- Perform other duties as assigned. (5%)
Department Staff: The holder of the position will have continuous contact with a research assistant. The incumbent will manage/guide research assistants with conducting experiments, will assign experimental tasks, will resolve questions and correct scientific errors, and, if required, will recommend disciplinary action. (50%)
University Faculty: The holder of the position will have moderate contact with SPORE PI and will be expected to provide information on the status of research projects, the status of submitted manuscripts, and own funding applications. The holder of the position will have moderate contact with entire SPORE faculty team to update them on research project status. The holder of the position will have moderate contact with the director of the National Center for Regenerative Medicine to coordinate collaborative research. (40%)
Students: The holder of the position may have continuous contact with Graduate Students. He or she may manage a student in developing own research projects to further goals of the overall research study. He or she may also supervise and evaluate graduate students who rotate through the laboratory. (10%)
The holder of the position will have supervisory responsibility including recommending reward or discipline of individuals who he or she will have responsibility for directing, including a research team consisting of graduate student and research assistant.
Education: Position will require a doctoral degree in the biomedical sciences plus experience as listed below.
Experience: Specific experience in murine models of GI diseases will be an asset.
EXISTING SKILLS or ABILITY TO LEARN THESE SKILLS
- Experience in mouse models of drug activity, or ability to learn this discipline.
- Experience in methods of protein preparation, ELISA assays, fluorometric assays, and assay of cell populations, or ability to learn this discipline.
- Knowledge and experience with working with drug inhibitors of the 15-PGDH pathway, or ability to learn this discipline.
- Demonstrated experience in writing and submission of scientific manuscripts.
- Ability to work collaboratively in studies requiring coordination between the Case GI SPORE and the National Center for Regenerative Medicine.
Working conditions will require working at the bench in a molecular genetics/ biology laboratory, working with mice and other animal models in the laboratory and in the animal facility, and, when needed, working outside standard working days or hours as required by the needs of a given experiment.
