X-ray Absorption Spectroscopy uses the tunable broadband X-ray radiation delivered by synchrotrons to provide electronic and structural parameters for the metal-containing active sites of metalloproteins, which comprise >30% of all proteins. This includes determination of bond lengths to very high precision (to ±0.02 Å), identification of metal oxidation state and electronic site symmetry, and identification of metal content in candidate metalloproteins via metalloproteomics screening methods. XAS can be performed on materials in a variety of states, including liquids, solids, and single crystals. The X-ray Spectroscopy Core provides access to scientific expertise and state-of-the-art resources for biological XAS, located at Brookhaven National Laboratory's NSLS and NSLS-II synchrotrons.
