One major goal of my group is to obtain mechanistic understanding of growth factor signaling, and the implications of these findings to aberrant cell functions in the setting of different diseases. Specifically, we are studying mitogenic signaling pathways that are regulated by small GTPases of the Rho family and their upstream activators, guanine nucleotide exchange factors (GEFs) from the Dbl family, which mediate and control signaling information flow from cell surface receptors to the intracellular machinery. In this area, we focused our efforts on both the GTPases themselves and their upstream GEFs, and their actions in normal and malignant settings. In addition, we devote a major research effort to the roles and molecular mechanisms that regulate reactive oxygen species in the context of human health and disease, and the impact of antioxidants (e.g. vitamin E) levels in the context of pathologies such as neurodegeneration, cancer, and metabolic perturbations. Our approach in attacking both of these fundamental questions is highly interdisciplinary and relies on diverse experimental approaches, including molecular and cell biology, biophysics, synthetic chemistry, and high-resolution microscopy. In addition, our work has important translational aspects in that we examine the relevance of our findings to human disease through collaborations with epidemiologists, geneticists and clinicians.
